NMN is a compound the body can use to make NAD+, a molecule involved in cell energy and other processes. Human studies using 250 to 1,000 milligrams a day reliably raise blood NAD+ and generally report good short-term tolerance. Reported benefits are modest, including better aerobic capacity and muscle function in some older adults, and lower blood pressure in some people with metabolic syndrome, a combination of health problems such as high blood pressure and blood sugar. A newer large trial reported improved cognitive function, meaning thinking and memory, but more research is needed to confirm how meaningful this is. Product quality varies, and NMN’s status as a supplement is legally uncertain in the United States.
What this means for you: NMN reliably raises NAD+, but evidence that it slows aging or produces large health improvements is still limited. Treat it as an experimental supplement and check product testing and local regulations.
moderate evidenceNMN sits between NR (nicotinamide riboside) and NAD+ in the salvage pathway. Whether it is superior to NR is the field's most public argument, largely a debate between David Sinclair's camp (NMN) and Charles Brenner's camp (NR). Both parties have commercial interests.
The scientific facts: NMN raises blood NAD+ in humans across multiple trials, at doses of 250-1000 mg/day. It is well-tolerated. Direct comparison studies against NR are rare and small. The claim that NMN is transported into cells intact via the Slc12a8 transporter is disputed. The competing claim, that NMN is dephosphorylated to NR before entering cells, is well-supported.
Human clinical benefits reported: modest improvements in aerobic capacity in older Japanese men, mild improvements in muscle function, some blood pressure reductions in metabolic syndrome. None of these are large effects.
Regulatory status matters. The US FDA excluded NMN from the supplement definition in 2022 because it had been investigated as a drug first, then lifted that exclusion on 29 September 2025. NMN is sold as a dietary supplement again in the US. Most other jurisdictions never restricted it. The FDA has not objected to the ingredient itself, but no NMN product carries an FDA approval, because supplements never do.
Quality varies. Third-party testing has repeatedly found NMN products significantly underdosed relative to their label claim. Look for products with a public Certificate of Analysis.
2026 Clinical Trial Update: First Large-Scale NMN RCT
The largest randomized, double-blind, placebo-controlled trial of NMN to date reported significant improvement in cognitive function [1]. Prior NMN human trials were small (n=20-80) and focused on physical endpoints like grip strength, walking speed, and insulin sensitivity. This trial extends the evidence base to cognitive outcomes, which are the endpoint most consumers care about.
A separate Nestle Health Science-led study found that 14-day supplementation of NR and NMN combined altered metabolic health markers in ways not previously described [2]. The study is industry-funded and not yet peer-reviewed, but it suggests NAD+ precursors may have metabolic effects beyond simple NAD+ elevation.
Early-phase trials of NAD+ precursors in Alzheimer's patients have also reported improvements in memory, physical movement, and metabolic health [3]. These are small, early-stage trials and should not be interpreted as evidence that NAD+ supplements treat or prevent Alzheimer's.
NMN Regulatory Context
NMN faces regulatory uncertainty in the US after FDA's 2022 determination that NMN cannot be marketed as a dietary supplement due to its prior investigation as a drug. The EU has not made a similar determination. Consumers should be aware that NMN availability may change based on regulatory decisions in different jurisdictions.
