ADD-IT1: A Randomized Study of Adjuncts to Immunotherapy in Patients With Advanced Cancers (Phase 2)
Phase 2 trial will test propranolol, desloratadine and magnesium plus vitamin D as adjuncts to cancer immunotherapy in 46 patients, University of Southern California. Starts November 2026. Not yet recruiting.
NCT07836712 (ADD-IT1) will randomise 46 patients with advanced cancers to standard immunotherapy with added adjuncts: propranolol, desloratadine, and magnesium plus vitamin D supplements. The sponsor is the University of Southern California. Phase 2, not yet recruiting, planned start 1 November 2026, primary completion November 2027.
Magnesium is one arm component among several adjuncts, not the tested intervention on its own. Any positive result would support a role for magnesium status in immune checkpoint inhibitor response, not magnesium supplementation for healthy adults.
The trial is registered but has not started. A registered trial with no results never moves an evidence grade. Magnesium stays at evidence grade A on the strength of its existing human trial base.

Multi-arm adjunct design means magnesium effects cannot be isolated from propranolol and desloratadine. Oncology population, not a healthy-aging population.
