What it is

Your cells run on a molecule called NAD+. It powers energy production and DNA repair. NAD+ drops as we age. NMN is a compound your body converts into NAD+.

Why people take it

The theory: if you replenish NAD+, cells work more like they did when you were younger. Blood NAD+ does go up when you take NMN. Whether that translates to living longer or healthier is still an open question.

Our verdict

The biology is real. The evidence for actual longevity or muscle/cognitive benefits in humans is thinner than the marketing suggests. Well-studied for safety at 250 to 2000 mg/day; efficacy band 250 to 900 mg/day.

Structure & class

β-Nicotinamide Mononucleotide (β-NMN). Intermediate in the NAD+ salvage pathway, one enzymatic step from NAD+ via NMNAT.

Mechanism & clinical data

Human RCTs (Yoshino 2021; Yi 2023; Liao 2021) confirm dose-dependent increases in blood NAD+ and PBMC NAD+. Efficacy signals appear from 250 to 300 mg upward, strengthen to about 600 mg, and flatten between 600 and 900 mg with no demonstrated clinical advantage above 900 mg (Yi 2023 head-to-head; Zhang 2026 meta-analysis dose subgroup p=0.9344). Endpoint benefits (VO2max, aerobic capacity, gait speed, insulin sensitivity) reported in some trials but effect sizes small and populations narrow.

Pharmacokinetics

Rapid oral absorption; NAD+ half-life in tissue 5 to 7 hours.

Dose & forms

500 mg
/ day
Range in trials

What to watch out for

Absorption varies wildly by form. Sublingual and liposomal claim better uptake but human head-to-head data is limited.

Safety limits and interactions

When a product exceeds the UL or contains an ingredient with clinically significant interactions, a "Review advised" flag appears on its product page and links here.

Safety note
No formal UL established by NIH/EFSA/FNB; long-term human safety data are still limited. Human trials show good tolerability up to 1,250 mg/day for 4 weeks, and EFSA's 2025/2026 novel food opinion approved NMN supplement use up to 300 mg/day.

Where this dose comes from

The trial dose above is our editorial pick from the published human trials for this ingredient. We choose the dose that the best-available studies used on the endpoint most relevant to longevity. The range shows the lowest and highest doses tested in published trials. The landmark studies below are the papers we read to set it. You can click through and check our work.

When new trials publish, our automated sweeps flag them. If a new study materially changes the dose, we update it through editorial review and record the old dose, new dose, and the triggering study in every affected product's recommendation history. The dose is never changed automatically.

Regulatory status

EUUnauthorised Novel Food; EFSA positive safety opinion adopted 4 March 2026 at up to 300 mg/day (β-NMN, EffePharm dossier). Commission listing pending.
USFDA reversed exclusion 29 September 2025; NDIN 1247 and NDIN 1259 reinstated 2 December 2025; NDIN 1444 (β-NMN, Effepharm) response 28 January 2026. NMN is a dietary ingredient again; each marketer still needs NDIN coverage.
UKNot authorised as Novel Food.
AUTGA permits up to 500 mg/day for 12 weeks or less in listed medicines from 10 December 2025 (SyncoZymes sponsor until 10 December 2027).
CANot permitted as NHP.
CNNot approved as food additive, new food raw material, drug or health food; cross-border e-commerce is the only legal import route.

Timeline

Regulatory events, evidence grade changes, and key research milestones for NMN, most recent first. Studies link to the paper; regulatory entries name and link the responsible authority.

  • 2026-07 Regulatory unverified
    Reported FDA reversal, unverified
A source reported that the FDA once again pulled its supplement-status ruling on NMN. NO1GEVITY has not been able to confirm this reversal with a primary source and treats it as unverified pending confirmation.
  • 2026 Research milestone
Zhang et al. 2026 meta-analysis (Nutrients)
A meta-analysis of 10 RCTs (349 participants) found NMN supplementation significantly reduced both systolic and diastolic blood pressure in adults with elevated blood pressure, with the effect most pronounced in hypertensive subgroups.
  • 2025-12 Regulatory confirmed
FDA re-affirms NMN's legal supplement status
In a follow-up statement, the FDA re-confirmed that NMN was legally marketable as a dietary supplement, ending most of the market uncertainty for US retailers.
  • 2025-09 Regulatory confirmed
FDA reverses position, NMN can be marketed as a supplement
After industry pressure and clarifying legal action, the FDA reversed its earlier stance and allowed NMN to be sold as a dietary supplement in the United States.
  • 2025-04 Regulatory single source
EU Court sharpens the bar for botanical health claims
A Court of Justice of the European Union ruling in April 2025 further tightened the requirements for how botanical ingredients (herbs, plant extracts) can be marketed with health claims in the EU, effectively removing much of the flexibility of the on-hold list.
  • 2024-06 Regulatory confirmed
EU report identifies 117 supplement substances as potential health risks
The HoA Working Group on Food Supplements (26 European countries) published its first report evaluating 117 substances used in food supplements. Of these, 65 were assessed as likely novel foods, 6 were already regulated, 34 had insufficient data, and 12 were prioritized for Article 8 restriction under Regulation (EC) No 1925/2006. The 12 priority substances include ashwagandha, curcumin, melatonin, piperine, St. John's wort, coumarin, p-synephrine, maca, holy basil, Melaleuca, black cohosh, and L-tryptophan. The report drew on 1,500 RASFF alerts reviewed between 2017 and mid-2022.
  • 2024 Regulatory single source
Health Canada, NMN requires product licence (NPN)
Health Canada regulates NMN under the Natural Health Products Regulations; a product needs an NPN to be sold legally. Very few NMN products carry an NPN, so most are technically not authorised for sale in Canada.
  • 2024 Research milestone
    NMN meta-analysis 2024 (12 RCTs)
A meta-analysis of 12 RCTs (513 participants) found NMN reliably raises blood NAD+ but shows no significant effect on most glucose or lipid outcomes, suggesting metabolic benefits may be overstated.
  • 2023-01 Regulatory single source
UK regulator queries NMN under Novel Food rules
The UK Food Standards Agency (post-Brexit) began treating NMN as an unauthorised novel food, echoing the EU position, though enforcement in practice has remained inconsistent and .com sellers continue to ship to UK consumers.
  • 2023 Research milestone
    Yi et al. 2023 (GeroScience)
In an 80-person, 60-day dose-dependent RCT, oral NMN (up to 900 mg/day) significantly raised blood NAD+ levels, was safe and well-tolerated, and improved six-minute walking distance versus placebo.
  • 2022-11 Regulatory confirmed
FDA excludes NMN from the definition of a dietary supplement
The FDA notified two companies that NMN was excluded from the supplement definition under the Federal Food, Drug, and Cosmetic Act because a new drug application referencing NMN preceded any legitimate marketing as a supplement. This triggered market confusion in the US.
  • 2022 Research milestone
Okabe et al. 2022 (Frontiers in Nutrition)
A 30-person, 12-week RCT found 250 mg/day oral NMN significantly increased whole-blood NAD+ levels with no adverse effects.
  • 2022 Research milestone
    Uthever RCT 2022
In 66 adults aged 40-65 over 60 days, NMN (300 mg/day) increased serum NAD+/NADH by 38% versus baseline and improved SF-36 quality-of-life scores compared to placebo.
  • 2006-12 Regulatory confirmed
    EU Health Claims Regulation adopted
Regulation (EC) No 1924/2006 laid down harmonised rules for nutrition and health claims made on foods, including supplements. Only claims listed in the EU Register of authorised claims can be used. Rejected botanical claims are held in a separate on-hold list.
  • 1997 Regulatory confirmed
EU introduces the Novel Food Regulation
Regulation (EC) No 258/97 required any food or food ingredient without a history of consumption in the EU before 15 May 1997 to be authorised before being placed on the market. This is the framework that captures NMN, spermidine as a supplement ingredient, urolithin A, and many other longevity actives.

Best-supported claim

NMN reliably raises blood NAD+ levels in humans. A 2024 meta-analysis of 12 RCTs (513 participants) by Jiaqi Zhang and colleagues confirmed a significant rise in blood NAD after supplementation (https://pubmed.ncbi.nlm.nih.gov/39116016/). A separate 2024 meta-analysis of 9 studies (412 participants) by Jian-Peng Wang and colleagues found NMN improved gait speed (SMD 0.34 m/s, p=0.033) and lowered ALT in middle-aged and elderly adults (https://pubmed.ncbi.nlm.nih.gov/39185644/). These are the two strongest signals in the human data.

Not supported by the evidence base

Marketers describe NMN as a reversal agent for aging, promising restored energy, sharper cognition, and extended lifespan. Human RCTs do not support this. Zhang's 2024 meta-analysis found most clinically relevant outcomes, including fasting glucose, triglycerides, and cholesterol, showed no significant difference from placebo (https://pubmed.ncbi.nlm.nih.gov/39116016/). A 2025 review by Aisyah Kuerec and colleagues on muscle mass and function in adults over 60 found NMN and NR gave no significant improvement in skeletal muscle index, grip strength, knee extension, or short physical performance battery scores (https://pmc.ncbi.nlm.nih.gov/articles/PMC12022230/). No human trial has measured lifespan extension.

Typical dose

250-900 mg once daily, with trial doses ranging from 250 mg to 2000 mg per day

Safety

NMN is well tolerated across trial doses of 250-2000 mg/day for up to 24 weeks. A 2026 meta-analysis of 15 RCTs found no increase in overall adverse events, serious adverse events, or liver enzyme elevations (ALT/AST) versus placebo (https://pubmed.ncbi.nlm.nih.gov/42514320/). Long-term safety beyond 6 months has not been tested in humans. NMN is not FDA-approved as a dietary ingredient in the US and its legal status has shifted amid ongoing FDA review.

Frequently asked questions

What is NMN?

Your cells run on a molecule called NAD+. It powers energy production and DNA repair. NAD+ drops as we age. NMN is a compound your body converts into NAD+.

What dose of NMN should I take?

The trial dose in published human studies is 500 mg per day, with a range of 250 to 900 mg per day. Always consult a qualified clinician before starting any supplement.

Is NMN safe?

No formal UL established by NIH/EFSA/FNB; long-term human safety data are still limited. Human trials show good tolerability up to 1,250 mg/day for 4 weeks, and EFSA's 2025/2026 novel food opinion approved NMN supplement use up to 300 mg/day.

What is the best NMN supplement?

We track 39 products containing NMN. The top-ranked product scores 8.3 out of 10 on our composite scale, which evaluates dose, evidence, price, third-party testing, and transparency. See the comparison table above. Scores are editorial judgments, not a scientific standard.

What is the evidence grade for NMN?

NMN has evidence grade B, meaning it is supported by human RCTs. Grade A means replicated human RCTs with hard endpoints. Grade B means human RCTs with biomarker outcomes. Grade C means mixed or small human trials. See our evidence grading methodology.
