What it is

Another route to raise NAD+, the same energy/repair molecule NMN targets. NR is one step further away from NAD+ than NMN, but it's the only one of the two that's been formally approved in the EU as safe.

Why people take it

NR raises NAD+ reliably in humans. Multiple 8 to 12 week studies show modest improvements in some biomarkers. Whether it makes you live longer is unproven, but for staying at status quo it has more safety data than almost any longevity ingredient.

Our verdict

The safer, more boring cousin to NMN. If you want a NAD+ booster and care about approvals, NR is the pragmatic choice.

Structure & class

Nicotinamide Riboside Chloride (branded as Niagen). Salvage pathway intermediate, converts to NMN via NRK1/2 then to NAD+.

Mechanism & clinical data

Multiple RCTs (Martens 2018, Dellinger 2017, Elhassan 2019) demonstrate 40 to 90% increase in blood NAD+ at 300 to 1000 mg/day. Effects on blood pressure, arterial stiffness, and inflammation modest but reproducible.

Pharmacokinetics

Rapid absorption; NAD+ elevation persists 8+ hours.

What to watch out for

Almost all commercial NR uses licensed Niagen® from ChromaDex. If a product doesn't say Niagen, ask why.

Safety limits and interactions

When a product exceeds the UL or contains an ingredient with clinically significant interactions, a "Review advised" flag appears on its product page and links here.

Safety note
No formal UL established. EFSA concluded NR is safe for healthy adults up to 300 mg/day (230 mg/day for pregnant/lactating women); trials have used doses up to 2 g/day without serious adverse events.

Where this dose comes from

The trial dose above is our editorial pick from the published human trials for this ingredient. We choose the dose that the best-available studies used on the endpoint most relevant to longevity. The range shows the lowest and highest doses tested in published trials. The landmark studies below are the papers we read to set it. You can click through and check our work.

When new trials publish, our automated sweeps flag them. If a new study materially changes the dose, we update it through editorial review and record the old dose, new dose, and the triggering study in every affected product's recommendation history. The dose is never changed automatically.

Timeline

Regulatory events, evidence grade changes, and key research milestones for NR, most recent first. Studies link to the paper; regulatory entries name and link the responsible authority.

  • 2025-04 Regulatory single source
EU Court sharpens the bar for botanical health claims
A Court of Justice of the European Union ruling in April 2025 further tightened the requirements for how botanical ingredients (herbs, plant extracts) can be marketed with health claims in the EU, effectively removing much of the flexibility of the on-hold list.
  • 2024-06 Regulatory confirmed
EU report identifies 117 supplement substances as potential health risks
The HoA Working Group on Food Supplements (26 European countries) published its first report evaluating 117 substances used in food supplements. Of these, 65 were assessed as likely novel foods, 6 were already regulated, 34 had insufficient data, and 12 were prioritized for Article 8 restriction under Regulation (EC) No 1925/2006. The 12 priority substances include ashwagandha, curcumin, melatonin, piperine, St. John's wort, coumarin, p-synephrine, maca, holy basil, Melaleuca, black cohosh, and L-tryptophan. The report drew on 1,500 RASFF alerts reviewed between 2017 and mid-2022.
  • 2018 Research milestone
    Martens et al. 2018
6 weeks of 500 mg twice daily lowered systolic blood pressure in older adults with elevated BP.
  • 2017 Research milestone
    Dellinger et al. 2017
    8 weeks at 100 to 1000 mg/day dose-dependently raised NAD+ in blood.
  • 2006-12 Regulatory confirmed
    EU Health Claims Regulation adopted
Regulation (EC) No 1924/2006 laid down harmonised rules for nutrition and health claims made on foods, including supplements. Only claims listed in the EU Register of authorised claims can be used. Rejected botanical claims are held in a separate on-hold list.
  • 1997 Regulatory confirmed
EU introduces the Novel Food Regulation
Regulation (EC) No 258/97 required any food or food ingredient without a history of consumption in the EU before 15 May 1997 to be authorised before being placed on the market. This is the framework that captures NMN, spermidine as a supplement ingredient, urolithin A, and many other longevity actives.

2026 evidence refresh

Nicotinamide riboside raises blood NAD+ 2 to 3-fold but shows little effect on cognition, muscle, or blood pressure.

NR at 2000 mg/day raised NAD+ 2.6 to 3.1-fold but did not significantly improve cognition, fatigue, sleep, or mood versus placebo in long-COVID patients. source

Best-supported claim

NR consistently and reliably raises blood NAD+ levels, typically 2 to 3-fold, across nearly every human trial regardless of population or dose. Chao-Yi Wu and Edmarie Guzman-Velez's 2025 trial in long-COVID patients confirmed this pharmacodynamic effect using 2000 mg/day (https://pubmed.ncbi.nlm.nih.gov/41357333/). Yasir Elhassan and colleagues showed in 2019 that NR also reaches skeletal muscle in aged men, elevating the NAD+ metabolome and lowering circulating inflammatory cytokines after 21 days at 1000 mg/day (https://pubmed.ncbi.nlm.nih.gov/31412242/). This biomarker effect is the most reproducible finding in the NR literature.

Not supported by the evidence base

Marketers sell NR as an anti-aging, cognition-boosting, blood-pressure-lowering supplement. Human trials tell a different story on downstream effects. Miranda Orr's 2023 randomized trial in older adults with mild cognitive impairment found NR raised NAD+ 2.6-fold but produced no improvement on the Montreal Cognitive Assessment or other cognitive tests over 10 weeks (https://pubmed.ncbi.nlm.nih.gov/37994989/). The 2025 long-COVID trial found no significant benefit on cognition, fatigue, sleep, or mood despite the same NAD+ boost (https://pubmed.ncbi.nlm.nih.gov/41357333/). A 2025 pilot trial combining NR with exercise in hypertensive older adults found NR plus exercise was not superior to exercise alone for lowering systolic blood pressure (https://pubmed.ncbi.nlm.nih.gov/40770531/). Raising a biomarker is not the same as changing an outcome that matters.

Typical dose

300-1000 mg once daily, with some trials using up to 2000 mg/day

Safety

NR is well tolerated at doses up to 2000 mg/day for up to 24 weeks across multiple RCTs. The NR-SAFE trial in Parkinson's disease patients found high-dose NR safe but flagged it can accumulate certain metabolites that warrant monitoring in some populations. The 2025 long-COVID trial reported one serious adverse event, deemed unrelated to NR, and otherwise only mild effects like bruising or rash (https://pubmed.ncbi.nlm.nih.gov/41357333/). Long-term safety data beyond 6 months in healthy adults is limited.

Frequently asked questions

What is NR?

Another route to raise NAD+, the same energy/repair molecule NMN targets. NR is one step further away from NAD+ than NMN, but it's the only one of the two that's been formally approved in the EU as safe.

What dose of NR should I take?

The trial dose in published human studies is 300 mg per day, with a range of 100 to 1000 mg per day. Always consult a qualified clinician before starting any supplement.

Is NR safe?

No formal UL established. EFSA concluded NR is safe for healthy adults up to 300 mg/day (230 mg/day for pregnant/lactating women); trials have used doses up to 2 g/day without serious adverse events.
