What it is

A molecule your cells make and that's also in aged cheese, wheat germ, and soy. It triggers autophagy, the cellular recycling process that clears out damaged parts.

Why people take it

Observational studies link high spermidine intake to lower mortality. The largest human trial, SmartAge (2022), tested 0.9 mg/day for 12 months in 100 older adults and found no cognitive benefit versus placebo. Larger trials are in progress.

Our verdict

One of the more promising 'gerotherapeutics' with human data. Wheat-germ extract products vary widely in actual spermidine content, insist on assay-verified milligrams.

Structure & class

Symmetric polyamine (N-(3-aminopropyl)-1,4-butanediamine). Induces autophagy via EP300 inhibition; hypusinates eIF5A.

Mechanism & clinical data

Kiechl et al. 2018 (Am J Clin Nutr) showed dietary spermidine inversely associated with all-cause mortality. SmartAge (Schwarz et al. 2022, JAMA Network Open) found no change in mnemonic discrimination at 0.9 mg/day for 12 months in adults with subjective cognitive decline (difference -0.03, 95% CI -0.11 to 0.05, P=.47).

Pharmacokinetics

Rapid absorption; polyamine pool has slow turnover.

Dose & forms

Trial dose
1.2 mg
/ day
Range in trials
0.9 to 6 mg

Common forms
Wheat-germ extract capsule, Chlorella-derived, Pure synthetic

What to watch out for

Product labels often say 'wheat germ extract 800 mg', that could be 0.8 mg or 8 mg of actual spermidine. Insist on labeled spermidine content.

Safety limits and interactions

When a product exceeds the UL or contains an ingredient with clinically significant interactions, a "Review advised" flag appears on its product page and links here.

Safety note
No formal UL (not an essential nutrient); short-term trials show no adverse effects at doses up to 40 mg/day for 28 days, well above typical dietary/supplemental intake (~6 mg/day).

Where this dose comes from

The trial dose above is our editorial pick from the published human trials for this ingredient. We choose the dose that the best-available studies used on the endpoint most relevant to longevity. The range shows the lowest and highest doses tested in published trials. The landmark studies below are the papers we read to set it. You can click through and check our work.

When new trials publish, our automated sweeps flag them. If a new study materially changes the dose, we update it through editorial review and record the old dose, new dose, and the triggering study in every affected product's recommendation history. The dose is never changed automatically.

Timeline

Regulatory events, evidence grade changes, and key research milestones for Spermidine, most recent first. Studies link to the paper; regulatory entries name and link the responsible authority.

  • 2026-09-03 Evidence grade
    Evidence grade changed: B → C
Downgraded after fact-check of study records. SmartAge (Schwarz 2022, N=100, JAMA Netw Open) is the largest and longest RCT of spermidine for cognition and missed its primary endpoint (mnemonic discrimination difference -0.03, 95% CI -0.11 to 0.05, P=0.47). Only supportive human data is Wirth 2018 pilot (N=30, CI crossed zero). A fabricated citation was removed from wiki/content/studies/. Grade B was not supported.
  • 2026 Research milestone
Jiang et al. 2026 (npj Aging)
Comprehensive review identifying eIF5A-TFEB axis as central autophagy mechanism. SmartAge Phase IIb showed no memory improvement at 12 months. 3.3 mg/day rice germ extract improved cardiometabolic biomarkers. 40 mg/day high-purity spermidine safe for 28 days in older men.
  • 2025-04 Regulatory single source
EU Court sharpens the bar for botanical health claims
A Court of Justice of the European Union ruling in April 2025 further tightened the requirements for how botanical ingredients (herbs, plant extracts) can be marketed with health claims in the EU, effectively removing much of the flexibility of the on-hold list.
  • 2024-06 Regulatory confirmed
EU report identifies 117 supplement substances as potential health risks
The HoA Working Group on Food Supplements (26 European countries) published its first report evaluating 117 substances used in food supplements. Of these, 65 were assessed as likely novel foods, 6 were already regulated, 34 had insufficient data, and 12 were prioritized for Article 8 restriction under Regulation (EC) No 1925/2006. The 12 priority substances include ashwagandha, curcumin, melatonin, piperine, St. John's wort, coumarin, p-synephrine, maca, holy basil, Melaleuca, black cohosh, and L-tryptophan. The report drew on 1,500 RASFF alerts reviewed between 2017 and mid-2022.
  • 2022 Research milestone
SmartAge (Schwarz et al. 2022, JAMA Network Open)
12-month RCT in 100 adults with subjective cognitive decline: 0.9 mg/day spermidine did not improve memory or biomarkers versus placebo.
  • 2020 Regulatory confirmed
Wheat-germ-derived spermidine allowed as novel food (limits apply)
Spermidine-rich wheat-germ extract (Longevity Labs' spermidineLIFE) was authorised as a novel food in the EU with maximum daily intake limits, allowing legal EU sales of spermidine-based supplements.
  • 2019-01 Regulatory confirmed
Wheat-germ-derived spermidine allowed as novel food (limits apply)
Wheat-germ extract containing spermidine was permitted as a traditional food ingredient. Concentrated spermidine supplements exceeding 7 mg per daily portion require Novel Food authorization. This creates a split market: wheat-germ extracts are legal, pure spermidine concentrates are not.
  • 2006-12 Regulatory confirmed
    EU Health Claims Regulation adopted
Regulation (EC) No 1924/2006 laid down harmonised rules for nutrition and health claims made on foods, including supplements. Only claims listed in the EU Register of authorised claims can be used. Rejected botanical claims are held in a separate on-hold list.
  • 1997 Regulatory confirmed
EU introduces the Novel Food Regulation
Regulation (EC) No 258/97 required any food or food ingredient without a history of consumption in the EU before 15 May 1997 to be authorised before being placed on the market. This is the framework that captures NMN, spermidine as a supplement ingredient, urolithin A, and many other longevity actives.
2026 evidence refresh

Spermidine triggers autophagy in cell and animal models, but the largest human trial found no cognitive benefit.

12 months of spermidine supplementation, raising daily intake by about 10%, did not improve mnemonic discrimination memory versus placebo in older adults with subjective cognitive decline. source

Best-supported claim

The strongest human evidence for spermidine is a small positive signal, not a confirmed benefit. Anna-Katharina Schwarz's 2018 pilot trial found spermidine supplementation moderately improved mnemonic discrimination in 30 older adults over 3 months, Cohen's d of 0.79 (https://pubmed.ncbi.nlm.nih.gov/30388439/). This pilot result motivated a larger follow-up. That larger trial, the 100-person SmartAge study led by Anna Flöel and Christian Schwarz, ran for 12 months and found no significant benefit on the same primary memory measure (https://jamanetwork.com/journals/jamanetworkopen/fullarticle/2792725). The field's own confirmatory trial did not replicate the pilot signal.

Not supported by the evidence base

Marketers present spermidine as a proven cognitive and longevity supplement based on autophagy research in worms, flies, and mice. The best-designed, largest human trial to date, SmartAge, found 12 months of spermidine supplementation did not modify memory or other biomarkers in people at risk for Alzheimer's disease (https://jamanetwork.com/journals/jamanetworkopen/fullarticle/2792725). A 2023 pharmacokinetic study by Stefan Senekowitsch and colleagues raises a more basic problem: high-dose spermidine supplementation did not measurably increase spermidine levels in blood plasma or saliva of healthy adults, suggesting the body tightly regulates spermidine and oral supplements may not change tissue exposure the way marketing implies (https://pmc.ncbi.nlm.nih.gov/articles/PMC10143675/). No human trial has shown spermidine extends lifespan or reverses aging markers.

Typical dose

1-6 mg once daily as spermidine-rich plant extract, though pharmacokinetic data cast doubt on absorption

Safety

Spermidine appears safe at studied doses. Patrick Keohane and colleagues' 2024 trial of a novel high-purity spermidine supplement at 40 mg/day in 37 healthy older men found no significant changes in clinical, lipid, chemistry, or hematological parameters, and no product-related adverse events over 28 days (https://linkinghub.elsevier.com/retrieve/pii/S027153172400126X). Long-term safety beyond a year has not been established in a randomized trial.
